ABSTRACT Cell suspension culture has been used as a model to study metabolic changes to several stresses. To have detailed information of nitrogen (N) limitation on sugarcane metabolism, a controlled study of the primary metabolites and representative compounds of secondary metabolism was developed using suspension cells growing under three different N regimes: normal condition (40 mmol·L–1 NO-3), slightly deficient (12 mmol·L–1 NO-3) and completely deficient (0 mmol·L–1 NO-3). Sugarcane cells were harvested after 3 and 7 days of treatment. A range of changes in the levels of amino acids, organic acids, sugars and phenolic compounds were observed upon the growth conditions applied. Nitrogen limitation remarkably affected the amino acids and carbohydrates biosynthesis, which, associated with the changes observed on phenolic compounds contents, indicates the upregulation of carbon sink compensation mechanisms in these sugarcane cells exposed to N starvation. As expected, the results showed that N limitation might cause an extensive metabolic reprogramming of both carbon and N metabolism in sugarcane cells, and these changes are related to the intensity of the starvation. Nitrogen is essential for plant growth and development, and its limitation sharply reduces crop yield. Thus, these results open new perspectives for in planta studies concerning carbon and N metabolisms balance in this crop.

Plantas de mandioca (Manihot esculenta Crantz) apresentam redução no acúmulo de matéria seca quando cultivadas com altos níveis de amônio na solução do solo. A razão para esse efeito do amônio ainda permanece pouco estudada. O presente trabalho foi desenvolvido visando avaliar a influência do NO3- e do NH4+ sobre alguns aspectos relacionados à fotossíntese e à concentração de alguns compostos nitrogenados em plantas de mandioca. Para esse estudo, utilizou-se a variedade Cigana Preta, que foi cultivada em solução nutritiva, tendo areia grossa lavada como substrato. Os tratamentos consistiram de três proporções entre NO3-:NH4+, em mM (12:0, 6:6, e 0:12). Plantas cultivadas apenas com NO3- ou com NO3- + NH4+ não apresentaram diferenças na taxa fotosssintética. No entanto, observou-se redução desse evento quando o NH4+ foi a única fonte nitrogenada. O valor da condutância estomática obtido para as plantas cultivadas apenas com NO3- foi de 0,49mol m-2s-1, enquanto que para as plantas cultivadas apenas com NH4+, o valor foi de apenas 0,16mol m-2s-1 (três vezes menor). Não houve diferenças significativas na concentração foliar de amônio livre, sugerindo não ter ocorrido efeito tóxico direto do NH4+ sobre a fotossíntese. A concentração de proteínas foi maior para as plantas cultivadas apenas com NH4+. Os resultados sugerem que o menor acúmulo de matéria seca das plantas de mandioca cultivadas exclusivamente com NH4+ é decorrente, dentre outros fatores, do efeito desse íon sobre a atividade fotosssintética, em virtude, principalmente, de sua ação negativa sobre a condutância estomática.

Cassava (Manihot esculenta Crantz) plants present reduction in dry matter accumulation when grown at high levels of ammonium in the soil solution. The reason for that still remains to be determined. The present research was carried out in order to evaluate the influence of nitrate and ammonium on aspects related to photosynthesis and the concentration of nitrogen compounds of cassava plants. Cultivar Cigana Preta plants were grown in nutritive solution with sand as substrate. Treatments consisted of three ratios of NO3-:NH4+, in mM (12:0, 6:6, e 0:12). Plants grown only on NO3- or NO3- + NH4+ did not show differences in photosynthetic rates, but reduction of them were observed for NH4+ as the only nitrogen source. Stomatal conductance of plants grown on nitrate was 0.49mol m-2s-1 and three times less for plants grown only on ammonium (0.16mol m-2s-1). There were no significant differences in leaf concentration of free ammonium among treatments, suggesting no direct toxic effect of ammonium on photosynthesis. Protein concentration was higher for plants grown only on ammonium. The results strongly suggest that the lower dry matter accumulation of cassava plants grown just on ammonium is due, among other factors, to its effect on photosynthetic activity, mainly because of its negative influence on stomatal conductance.

Desenvolveu-se o presente estudo visando-se avaliar o efeito da fonte nitrogenada sobre o acúmulo de matéria seca e a absorção, distribuição e utilização de nitrogênio em mandioca. Para a instalação do experimento utilizou-se a variedade Cigana preta, cultivada em solução nutritiva e em vasos com capacidade para 11,0 L. O nível de nitrogênio na solução foi de 12 mol m-3 e os tratamentos consistiram de cinco proporções de NO3-:NH4+ , conforme segue: 12:0, 9:3, 6:6, 3:9 e 0:12 mol m-3. No encerramento do experimento, que ocorreu 90 dias após o plantio, verificou-se que as menores quantidades de massa seca total foram obtidas para as plantas cultivadas tendo apenas nitrato ou amônio como fonte de nitrogênio. Nesse aspecto, o amônio foi mais prejudicial ao crescimento do que a fonte nítrica. A concentração de NH4+ livre no tecido vegetal foi maior para as raízes de absorção, seguida pelas folhas e raízes de armazenamento. A eficiência de absorção de nitrogênio foi maior para as plantas cultivadas sob as maiores proporções de NH4+. Os resultados evidenciaram que a cultura da mandioca cresce melhor quando ocorrer na solução de crescimento uma mistura de NO3- e NH4+, principalmente nas proporções de 9:3 e 6:6 mol m-3.

This study was carried out to evaluate the effect of nitrogen source on dry mass accumulation and absorption, distribution and also use of nitrogen by cassava plants. Cigana Preta cultivar plants were grown in nutrient solutions set up in 11.0 L containers. Nitrogen concentration in solutions was 12 mol m-3. Treatments included five combinations of NO3-:NH4+ , that is, 12:0, 9:3, 6:6, 3:9 and 0:12 mol m-3. Aftert 90 days the dry mass was determined. The lowest total dry mass values were obtained for plants grown with just nitrate or ammonium as nitrogen source. In this sence, ammonium affected growth more than the nitric source. The concentration of free NH4+ was higher for absorption in roots, followed by leaves and storage roots. The efficiency of nitrogen absorption was higher for plants cultivated under highest proportions of NH4+. The results showed that cassava plants grow more when growth solution contains a mixture of NO3- and NH4+, especially at the proportions of 9:3 and 6:6 mol m-3.
